Outline of Research at the Start

ワクチンは人類を感染症の脅威から守る優れた医療戦略であるが、副作用がより少なく安心して使用できる次世代のワクチン開発が切望されている。
本研究では「アジュバントを使用しないワクチン」の実現可能性を視野に入れ、リンパ節濾胞樹状細胞(FDC)の機能増強に関わる細胞および分子レベルの新知見の取得を目的とする。具体的には、①FDC分化経路、②FDC分化に関わる細胞ネットワーク、③FDC分化誘導因子の同定を目指す。

Outline of Final Research Achievements

We succeeded in proving differentiation of FDCs from MRC in both lymph nodes and Peyer's patches using inducible RANKL(Tnfsf11)-fate mapping system. We also succeeded in establishing ILC3/LTi-specific decreased mouse model focusing on ILC3/LTi-specific enhancer region. However, relationship between FDC differentiation and LiT cell function was not clarified yet.

Academic Significance and Societal Importance of the Research Achievements

FDCは抗原特異的B 細胞の成熟に必須の線維芽細胞であるが、その由来や分化経路は不明な点が多い。本研究により、FDCの由来が MRCであることが明らかとなった。現在作出中のFDC特異的なレポーターマウスを活用することにより、FDC特異的な分子群や生体内における機能がより詳細に解明できる。また、これまでILC3/LTi細胞特異的な欠損マウスモデルが存在しなかったが、本研究により「二次リンパ組織形成には影響がなく、LTi細胞のみが特異的に減少したマウス」モデルの作出成功したため、成体リンパ組織におけるLTi細胞の機能解明が飛躍的に進むと期待される。
