Outline of Research at the Start

活性化による血小板細胞の機能変化は膜糖タンパクGPIIb/IIIaの細胞外ドメインの高次構造変化による。細胞内ドメイン・細胞膜貫通ドメイン・細胞外ドメインを統合したGPIIb/IIIaモデルを、分子を構成する全ての原子と周囲の水分子の位置座標から作成する。血小板細胞活性化に伴う細胞内環境のわずかな変化がGPIIb/IIIaの細胞外ドメインの大きな構造変化を惹起するメカニズムとして「分子レバレッジ機構」の解明を目指す。

Outline of Annual Research Achievements

血小板活性化時にリガンド蛋白との接着性が大きく変化する膜糖蛋白GPIIb/IIIaに注目し、細胞内ドメイン・膜貫通ドメイン・細胞外ドメインを連成したモデルを作成した。連成モデルの作成にはX線結晶構造解析、NMRなどと矛盾しないモデルを作成した。申請者が学内に保有するGPU 4基を実装したHPC5000-XSLGPU4TSにて膜糖蛋白GPIIb/IIIaの細胞内ドメイン・膜貫通ドメイン・細胞外ドメインを構成する全ての原子および周囲に配置した水分子の位置座標から初期構造を作成し, CHARMM力場を用いて各原子の位置座標と速度ベクトルを2フェムト秒ごとに計算した。当初の研究計画では膜糖蛋白GPIIb/IIIaの細胞内ドメイン・膜貫通ドメイン・細胞外ドメインを連成したモデルの安定構造を確定し、安定構造における細胞内ドメインのGPIIb Pro999と GPIIIa TYR747の重心間距離の算出を目指した。しかし、大規模計算を継続する過程にて、以前に申請者が解析した血小板膜糖蛋白GPIbalphaと異なり、GPIIb/IIIaの細胞内・膜貫通・細胞外ドメインの連成モデルの構造が安定しないことがわかった。特に、細胞内ドメインの構造は不安定であった。当初の計画通り、GPIIb Pro999とGPIIIa TYR747の重心間距離を80オングストロームに拘束すると計算は破綻した。GPIIb/IIIaの細胞外ドメイン、細胞内ドメインともに構造が不安定であり、分子レバレッジメカニズムは当初推定した仮説よりも複雑であることが解明された。GPIIb/IIIaの細胞内・膜貫通・細胞外ドメイン全体としての構造の不安定自体が、細胞活性化時の細胞内ドメインのわずかな高次構造変化と細胞外ドメインの大きな高次構造変化を惹起するメカニズムとして論文を作成した。
