Outline of Research at the Start

習慣的な運動は、生物時計が発振する生体リズムに影響することが知られている。マウスやラット等の実験動物を恒常環境で飼育した際に回転輪の有無により行動リズムのフリーラン周期が異なることが報告されている。しかし、運動から中枢時計の存在する視床下部視交叉上核へのフィードバック機構の存在およびその実態については依然として不明である。そこで、本研究では習慣的な運動が、生物時計中枢である視交叉上核内の細胞間リズムおよび視交叉上核外の脳部位との神経ネットワークに与える影響をin vitroおよび行動レベルで明らかにすることに挑戦する。本研究は、ヒトの生物時計システムのより一層の理解にもつながると期待している。

Outline of Annual Research Achievements

行動（睡眠・覚醒）および生体の多くの機能には、約24時間の概日リズム（サーカディアンリズム）が存在する。哺乳類におけるこのリズムの発振中枢は間脳視床下部視交叉上核に存在し、中枢時計と呼ばれています。生物時計の自律振動メカニズムは、複数の時計遺伝子＊3
の転写と翻訳からなる分子フィードバックループが想定されている。近年、生物発光技術の進展により、ホタルの発光酵素を導入したトランスジェニックマウスを用いることで、同一個体からの複数部位の時計遺伝子発現の測定が可能となった。本研究では、哺乳類の主要な時計遺伝子であるPeriod1を生物発光によりモニター可能なトランスジェニックマウス（Per1-luc マウス）を用いて、通常の昼夜変化に同調した状態と、昼夜変化の存在しない恒常暗環境、そして恒常暗環境下で24時間周期の運動スケジュールを与える3つの条件で、行動リズム、視交叉上核、弓状核、肝臓、骨格筋といった末梢組織の時計遺伝子発現リズムの時間関係を比較した。その結果、習慣的な運動は恒常暗下で行動リズム、SCN、末梢時計の位相関係が変化した後、行動リズム、中枢時計、末梢時計の時計遺伝子発現リズム間の時間関係を再統合し、昼夜変化による光同調と同様に行動リズムと中枢・末梢時計間の時間関係を維持することが可能であることを明らかにした。さらに、運動を行うタイミングに応じてマウスの活動時間が変化したことから、運動時刻に応じて行動リズムにおける活動開始を制御する Evening 振動体と活動終了を制御する Morning 振動体、2 つの振動体間の相互協調を変化させる可能性を新たに提示した。R4年度は、本研究成果を論文として発表した(Sato & Yamanaka Am J Physiol Regul Integr Comp Physiol 2023)。

Reason

本研究計画では、習慣的な運動が哺乳類の生物時計中枢である視交叉上核（SCN）に与える影響、生物時計の階層構造に与える影響を明らかにすることを目標としている。R4年度には、研究計画当初より行ってきた通常の明暗サイクル、恒常暗、そして恒常暗環境下で 24 時間周期の運動スケジュールを与えた3つの条件で、行動リズムとPer1-luc マウスから採取した SCN、弓状核、肝臓、骨格筋を培養し、時計遺伝子発現リズムを比較した実験データを解析し、行動リズムが恒常暗下での運動スケジュールに同調（非光同調）し、運動スケジュールに同調するタイミングが恒常暗下でのフリーラン周期に依存すること、活動時間の長さが昼夜変化の存在する条件に比べ恒常暗条件下で延長し、運動スケジュールに同調した際には再び短縮すること、運動スケジュールに同調した際の活動時間が運動のタイミングに依存して変化すること、活動の開始位相と終了位相では運動スケジュールへの同調に要する期間が異なることを明らかにした。さらに、行動リズムと視交叉上核、肝臓の時計遺伝子発現リズムの時間関係は、昼夜変化の存在する条件下から恒常暗条件下では異なっていたが、恒常暗下で運動スケジュールに同調した条件では昼夜変化の存在する条件と同様の時間関係に回復（再統合）されることを明らかにし、論文として発表した(Sato & Yamanaka Am J Physiol Regul Integr Comp Physiol 2023)。さらに、R4年度には高感度CCDカメラを用いて習慣的な運動がSCN内の細胞間ネットワークに与える影響を明らかにする実験、運動に対する行動リズムの位相反応には雌雄差に関する実験が終了し、データ解析を進めている。以上より、本研究計画はおおむね順調に進展しているといえる。

Strategy for Future Research Activity

R5年度は、本研究計画の最終年度であり、これまでに取得したデータの論文化を進めると共に、本研究計画により新たに明らかとなった運動の時刻依存的にSCN内の細胞間ネットワークが変化する可能性を検証するための新たな実験系を立ち上げる予定である。
