Outline of Research at the Start

細胞のエネルギー代謝に着目すると,がん細胞とアストロサイトは特殊な類似性をもっている。すなわち,がん細胞とアストロサイトは通常の細胞とは違い,好気的環境でも嫌気呼吸である解糖系を亢進させる。このような代謝特性から,両細胞が解糖系振動を起こすと期待される。本研究では,HeLa細胞以外のがん細胞に加え,アストロサイトの解糖系振動をはじめて観測することを目指している。
がん細胞とアストロサイトの解糖系振動と同期現象を対比させて研究することにより,生物学的な特徴や機能が解糖系という根源的な代謝系から議論できるものと期待される。本研究は,がんの早期発見や脳の機能障害の診断などの医学的な応用も期待される。

Outline of Final Research Achievements

We have studied glycolytic oscillations and their synchronization in cancer cells and astrocytes. We observed the oscillations in HeLa cervical, DU145 prostate, and MAD-MB-231 breast cancer cells in monolayers as wells as HeLa cells in spheroids. Adjacent HeLa cells in spheroids exhibited no synchronization as revealed by the order parameter for synchronization. Astrocytes exhibited spike-like behavior upon addition of glucose, however exhibited no sustained oscillations under some conditions such as addition of glutamate in the extracellular solution.

Academic Significance and Societal Importance of the Research Achievements

振動や同期は生命現象に広く見られ,重要な生物学的機能を有している。解糖系振動は細胞のエネルギー代謝過程でみられる細胞レベルの生物学的振動の一つであり,これまで酵母細胞や膵臓のβ細胞などで観察されてきた。本研究では,がん細胞も解糖系振動を起こすことを1細胞レベルではじめて明らかにした。がん細胞の種類や形態によって振動の特徴が異なることなどを明らかにした。特に,振動周波数が高いがん細胞ほど5年生存率や治療抵抗性などで判断される悪性度が高いことを示した。がん細胞と同様に解糖系を活発に利用するアストロサイトにおいても解糖系振動の特徴が明らかになれば脳機能との関連の解明につながることが期待される。
