Outline of Research at the Start

「3次元分子シミュレーションのような3次元動画を実際の分子で撮影できないか?」「有機分子に馴染みの薄い電子顕微鏡(電顕)の世界」を「電顕に馴染みの薄い有機化学の世界」に導入し,「分子電顕技術(molecular electron microscopy)」という新しい分子科学の手法を確立することでこの問いに答える.有機分子やその集合体の動きを3次元の動画により単一分子レベルでの絶対配置の決定,溶液中の極微量成分の一分子一分子の単離と構造決定,一つ一つの化学反応事象の「その場」観察というこれまで不可能と考えられてきた可能性を拓き,触媒,有機エレクトロニクス,生物科学に関わる基礎科学の革新を目指す.

Outline of Annual Research Achievements

2020年から21年にかけてGatan社のK3カメラを導入,画像取得条件とソフトウエアの最適化を行った.2022年度はこのカメラを活用して,「映像分子科学」研究の基盤を確立した.これまでに報告した,化学現象が機械的振動によって影響を受けるという現象は,これまでの化学の標準的な反応機構論の範疇を超えた現象である.この問題を精査するために,K3-ISカメラの高速性能を活かして,結晶の成長に対する振動の影響を原子レベルで調べた.その結果,CNTからの機械的刺激によってNaCl結晶状のNaClユニットが結晶面上を動き回りながら成長し,その後に最終的な落ち着き場所を見つけて結晶面の成長を引き起こすことが分かった.これらの観察を出発点として研究を発展させる準備を開始した.1934年に電顕が発明されて以来「電子線によって有機物はすぐに分解する」とされてきたが,分子がどのようにして電子線と化学的相互作用をするかに関する研究,すなわち放射化学の研究は近年余り大きな進展を見ていない.このことが電顕による分子の動的挙動の解明,すなわち「映像分子科学」の研究進展の足かせとなってきた.カーボンナノチューブの中に分子を詰めたり,表面に分子を化学的に固定して観察する我々独自の実験条件に対して,電子線結晶回折の条件では分子は結晶中の単位胞に固定されている.この2つの条件下での分子の挙動を比較研究することにより,電顕観察下での,電子線と有機分子の相互作用の本質の解明に着手した.これまでの研究により,カーボンナノチューブにつめたC60の反応は加速電圧に顕著な依存性を示し,ナノチューブの電子遷移とそこからのエネルギー移動によって化学反応が誘起されていると考えられる.一方で,電子線結晶構造解析条件下ではもっぱら振動励起が起きていることが分かった.

Reason

2021年から22年度の京急を経て,2019年に本研究を開始した当初は,単に分子の動きを見る,プロジェクトだったものが,当初計画になかった電子回折の速度論的解析という未踏の分野に踏み込むことができた.ここでは,有機結晶の電子回折のシグナル強度の減衰のキネティクスを測定して,減衰が一次速度式に乗ることを発見し,アレニウスプロットの頻度因子の中に分子と電子線の散乱断面積および結晶の融解エントロピーの情報が隠されていることが分かった.これは「電子線照射によって有機分子は直ぐさま分解する.」という1930年代からの常識を打ち破る発見である.

Strategy for Future Research Activity

電子回折データの中に隠された物質のエントロピー情報の発見は,化学熱力学と統計力学という19世紀以来独立の研究分野として発展してきた化学と物理の二つの分野を結びつける新しい展開である.今後,単に,見る,から統計力学方面への展開を図る.
