Outline of Research at the Start

世界人口を支える食糧生産には施肥は不可欠であるが、環境汚染や資源枯渇が問題である。作物は高い栄養吸収能を持つがその能力には限度があり、施肥は土壌の栄養濃度を高める。植物は栄養の乏しい自然環境での進化の過程で低栄養条件への適応能を獲得したが、適応には栄養濃度の感知が必須である。本研究は植物の無機栄養の感知機構を解明し、感知に伴う現象を統合的に解明し将来の食糧生産に資する知見を得ることを目的とする。

Outline of Annual Research Achievements

本研究では栄養感知の分子機構の解明と感知に伴って起こる現象の普遍的な解析を(1)細胞内、(2)細胞膜、(3)細胞壁の3つコンパートメントごとに進めきている。本年度の成果の概略を記す。(1)細胞内感知機構の構造的理解としてはホウ素依存的に起こるリボソームのAUGUAA上での停止後に起こるmRNA分解について、変異株から得られた原因遺伝子の一つがAUGUAAを持つmRNA上で停止したリボソームの周辺でmRNAを切断する可能性を調査するために、当該遺伝子がコードするタンパク質の断片を大腸菌で発現させ得られたタンパク質のRNA分解活性を調査したところ、分解活性があることが見出された。さらに当該変異株での分解されたmRNA断片の網羅解析を行ったところ、NIP5;1 mRNAの分解に影響を及ぼすことが確認された。また、ホウ素濃度を変化させたときの翻訳調節の網羅解析を行い、多くの遺伝子が翻訳制御を受けていることが明らかになった。 (2)細胞膜:細胞膜に局在するホウ素輸送体BOR1はホウ素濃度が高まると細胞内に選択的に取り込まれ分解される。BOR1と相互作用するタンパク質を解析するための各種系統の解析を行った。(3)細胞壁:ホウ素欠乏での生育が野生型よりも優れた複数のシロイヌナズナ変異株について、ペクチン含量等に異常があることが見出されている。本年度についても継続的に変異株について、原因遺伝子やその遺伝子産物の機能についての推定を進めた。
