Outline of Research at the Start

研究の目的であげた二つの目的を実行するため、光遺伝学を駆使してスパインレベルの機能イメージング法を開発し、個々のシナプスに入力する情報を可視化し、その変化を継続的に観察し、脳の情報処理の素過程と、学習・記憶の素過程をシナプスレベルで解明する。さらに、細胞集団のイメージングと組み合わせて、これらの素過程がネットワーク全体としての学習にどのように寄与しているのかを解明する。最後に、細胞集団を光遺伝学を用いて人工的に活性化し、細胞集団の活動が知覚・学習と因果関係を持つかどうかを検討する。

Outline of Annual Research Achievements

一次視覚野（V1）において、個々の神経細胞がどのような入力を受け取り、それを組み合わせてどのような出力を出しているのかその素過程の解明を試みた。Ｒ1年度に開発したin vivoで活動電位の逆伝播を停止させながら、スパインの２光子カルシウムイメージングを行う実験系を用い、視覚野の個々の興奮性ニューロンから、スパイン活動の大規模イメージングを行い、シナプス入力の空間機能マップを作製した。動物に視覚刺激を提示し、細胞体の活動を記録したのち、細胞体活動を光抑制しスパインの活動を２光子カルシウムイメージングにより記録した。大規模イメージングを行い、１個のニューロン当たり約1000個のスパインの活動を記録し、その結果を用いて、ニューロンの樹状突起上に約1000個のスパインの位置と反応を示し、シナプス入力の機能マップを作製した。さらに、このようにして作成したシナプス入力空間機能マップをもとに、シナプス統合のメカニズムを解析した。まず、細胞体と同じ反応を示すスパインが多いことがわかった。次に、細胞体と同じ反応をするこれらのスパインが、特定の枝にクラスターを形成していることがわかった。最後に、樹状突起統合モデルを立て、各スパインのカルシウムシグナル変化から細胞体の反応の予測が可能かどうか検証した。このとき、(1)スパイン入力の単純な加算、(2)スパインのクラスターを考慮に入れたモデル、(3)活動電位の発生の閾値を考慮したモデル等を検討したところ、(2)と(3)の両者を取り入れたモデルが、細胞体の活動を最も良く予測することがわかった。

Strategy for Future Research Activity

大脳皮質の神経細胞は、新規図形を学習し、それに対する反応性を獲得することができる。この学習はシナプスレベルで起こっていると考えられるが、実際にシナプスレベルでどのような変化が起こっているのか全くわかっていない。シナプスの学習則は、Hebb則やSTDPなど時間タイミングについての学習則として一部知られているが、細胞が担っている情報についての学習則が知られておらず、どのような学習則によって複雑な機能を持った細胞が出来てくるのか全く不明である。
今後の研究計画として、動物が新規図形を学習しているときの高次視覚野の神経細胞の反応性の変化を継時的に観察する。マウスに新規図形のペア（図形Ａ、Ｂ）を提示し、マウスの口先から少し話したところに水の吸い口を設置し、図形Ａを提示しているときに舌を出すと水が貰え、図形Ｂを提示しているときは貰えないというタスクを学習させる。このタスクを学習しているときの高次視覚野の神経細胞の反応性の変化を継時的に観察する。
