Outline of Research at the Start

マウス造血幹細胞の加齢特性をエピジェネティックな観点から明らかにする。特に、ストレスによるエピジェネティック変化の実態や、加齢とともに拡大する造血幹細胞の多様性のシングルセル解析などを行う。さらに、加齢ニッチ細胞の関与や、加齢に伴い著増する骨髄球系腫瘍の発症に関わる加齢造血幹細胞のエピゲノム要因を明らかにする。これらの解析を通して、加齢造血幹細胞の再活性化や形質転換の回避などの介入法を創出する。

Outline of Annual Research Achievements

若齢・加齢造血幹細胞のシングルセルRNA-seqを行ったところ、加齢造血幹細胞は若齢造血幹細胞とは異なるクラスターに細分化され、Clusterin (Clu) などの加齢造血幹細胞に特徴的な遺伝子群の発現が加齢とともに増強することが観察された。Clusterinは分泌型シャペロンとして機能する分子である。Cluの発現をGFPでマーキング可能なマウスを入手し、Clu発現幹細胞の挙動を解析したところ、造血幹細胞分画がClu陽性・陰性に大別できること、その比率が加齢とともに逆転することが明らかとなった。すなわち、若齢マウスではClu陰性造血幹細胞が大半を占めるが、加齢とともにClu陽性造血幹細胞が増加し、2年齢の加齢マウスでは約90％がClu陽性となる。Clu陽性造血幹細胞はMyeloid/plateletに偏った分化を示し、加齢マウスの血液細胞の特性を規定する。一方、Clu陰性造血幹細胞はリンパ球を含めてバランスの良い分化を示し、若齢造血幹細胞に近い表現系を持つ。RNA-seq解析によるトランスクリプトーム解析においては、上記の特徴が明瞭に示された。これらの2つの異なる造血幹細胞集団の割合の変化が血液の老化を大きく規定するものと考えられた。また、加齢関連骨髄性腫瘍として頻度の高い骨髄異形成症候群 (myelodysplastic syndromes : MDS) における転写制御ネットワークの異常を明らかにする目的で、MDS及び類縁疾患の幹細胞分画を用いてATAC-seqによるクロマチン特性解析を行った。その結果、MDS幹細胞分画のクロマチン状態は、前駆細胞分画に比べてより良く病態の多様性や悪性度を反映しており、またモチーフ解析の結果から、MDS幹細胞では造血幹細胞の機能に重要なHOX familyやAP1 family などの結合配列を含む領域が閉じる一方で、骨髄球系の分化に関わるCEBPやKLFなどを含む領域が開いており、正常な造血分化に関わる様々な転写ネットワークの異常が、MDSの多様性や予後を規定することが明らかになった。

造血幹細胞の加齢に伴う多様性の変化を明らかにすることができた。また、骨髄異形成症候群における病態の一端をその幹細胞と前駆細胞のクロマチン構造を解析することにより明らかにすることができた。ともに論文を準備中である。

Strategy for Future Research Activity

(1) 加齢造血幹細胞におけるClu陽性・陰性分画のATAC-seq解析やDNAメチル化解析を行い、これらの加齢造血幹細胞分画を制御する転写ネットワーク・エピゲノムの解析を進め、加齢造血幹細胞の機能低下に関わる分子基盤を明らかにする。
(2) 造血幹細胞の機能低下に関わるストレスシグナルの解明：1.で同定される造血幹細胞の機能低下に関わるクロマチン・エピゲノム特性が、どのようなストレスシグナルによって造血幹細胞に記憶されるのかについて、サイトカインの一つであるトロンボポイエチンがCluの発現制御に関わる予備的データを得ている。トロンボポイエチン欠損マウスやトロンボポイエチンチャレンジ試験などにより、加齢に伴う造血幹細胞の特性変化の機序の一端を明らかにする。
