Outline of Research at the Start

菌類ウイルスは,菌類の生理に影響を与えうるため,農作物の減収をもたらす植物病原糸状菌病をコントロールする上で重要な存在である.一方で菌類は,ウイルスに対する防御機構 (RNAi) を備えている.本研究では,菌類の抗ウイルスRNAi機構について,モデル糸状菌であるクリ胴枯病菌を用いて,分子レベルで解明することを目指す.具体的には,クリ胴枯病菌のRNAi機構において働く分子が,様々なウイルスの感染状況ごとに異なる可能性を検証する.

Outline of Annual Research Achievements

RNA干渉 (RNAiまたはRNAサイレンシング) 機構は,真核生物において広く抗ウイルス防御機構として働いている.本研究では,糸状菌-ウイルス間相互作用研究のモデル宿主であるクリ胴枯病菌を用いて,「Argoanute不要の抗ウイルスRNAi機構」の立証に取り組んできた.
(1) Argonaute不要のRNAiにDicerが必要であることの立証:
クリ胴枯病菌DK80系統を遺伝的背景としてdcl2および全てのagl遺伝子を欠損した変異株 (Δdcl2ΔaglQ) に,dcl2を相補することに成功した.本相補株 (Δdcl2ΔaglQ+dcl2) において,Argonaute不要の抗ウイルスRNAi機構が働くことが示唆されていたRNAウイルス [Rosellinia necatrix victorivirus 1 (RnVV1) およびmycoreovirus 2 (MyRV2)] の蓄積量を解析した.Δdcl2ΔaglQにおいて両ウイルスの蓄積量が顕著に上昇することとは対照的に,Δdcl2ΔaglQ+dcl2においてはDK80の場合と同様に両ウイルスがほとんど蓄積しなかった.このことから,Dicerを介したRnVV1やMyRV2に対する防御機構に,Argoanuteが寄与していないことが示された.すなわち,本研究の目的を達成することができた.現在,これらの成果をまとめた論文を執筆している.
Argonauteの抗ウイルス性がウイルスごとに異なる分子機構を解明するための実験計画(2)および(3)については,本年度は実験材料の作出ならびに予備実験の遂行に至った.
上記のプロジェクトと並行して,菌類ウイルスの性状やライフスタイルの多様性を広く明らかにするための研究にも取り組んできた.本年度はパキスタン・スイスの研究機関との共同研究により,キャプシドを持たない10分節RNAウイルスhadakavirus (2例目) や,複製酵素を分割してゲノムにコードする4分節RNAウイルスsplipalmivirus等の新規ウイルスの分子性状や生物学的特性を明らかにし,論文公開に至った.
