Outline of Research at the Start

脳の働きを担う神経細胞の超高解像解析を目指した新規イメージング基盤の開発を目指す。従来の多点電極アレイを用いた電気化学イメージング(可視化)法は神経伝達物質などの生体分子のリアルタイムな追跡が可能であるが、各電極点へのリード線の必要や微細加工技術の限界から得られるイメージの解像度は大きく制限されていた。
本研究ではリード線を必要としない"closedバイポーラ電極"を細密に並べた新規電極アレイを作製し、発光シグナルによる生体分子の超高解像度イメージングを目指す。この新規イメージング基盤の開発によって、これまでにない解像度で多くの神経変性疾患に関わる脳のエネルギー伝達機構の解明を目指す。

Outline of Annual Research Achievements

本研究では、生体分子のラベルフリーな高時空間分解能二次元イメージングを実現するバイポーラ電気化学顕微鏡の創成に関する研究を行っている。主に、①本顕微鏡システムの基板となる、多数の導電材料(バイポーラ電極)が絶縁基板を高密度に貫通した「バイポーラ電極アレイ」の作製方法の提案、②分析対象の拡張の2軸を基に検討を行ってきた。

当該年度は主に、導電ペーストを多孔基板に塗り込む手法によるバイポーラ電極アレイの作製方法の検討および最適化を行った。10ミクロン直径の孔が15ミクロンピッチで貫通した「ガラスキャピラリープレート」にカーボンペーストを塗り込み熱硬化させることで、高密度バイポーラ電極アレイの簡便な作製に成功した。本手法は、従来報告されてきたリソグラフィ技術を用いた作製方法とは異なり、特殊な装置を用いることなく安価、簡便にバイポーラ電極アレイの作製が可能であることが利点である。また、ペースト材料に触媒材料を混ぜ込むことで機能性の付与が可能であり、本研究では過酸化水素還元触媒であるPrussian blueを含有したカーボンペーストを用いることで過酸化水素の高時空間分解イメージングを達成した。

また、本顕微鏡システムにおいてドーパミンなどの還元体分子のイメージングを可能にする「Cathodicルミノフォア」の改良による、ドーパミン検出およびイメージングの高S/N化のための検討を行った。Cathodicルミノフォアとして前回用いたRu(bpy)32+/GSSGよりも低電位で電気化学発光を生じるN,N′-dimethyl-3,4,9,10-perylenedicarboximide (PDI-CH3)をバイポーラ電極アレイ上に修飾することで、S/N比を向上させたドーパミンの検出およびイメージングを実現した。
