Outline of Research at the Start

ヒトの腸管には約40兆個もの腸内細菌が生息している。近年、様々な疾患との関連が分かってきた。腸内細菌は個人差が大きく、多様性に富んでいることが知られている一方で、大腸がんに代表される疾患関連因子の探索において、その多様性が考慮されている例は少ない。これまでの研究で、国立がん研究センターで大腸内視鏡を受けた患者から糞便を収集し、メタゲノム解析を行った結果、大腸がんに関連のある菌種や代謝遺伝子などの多くの知見を得た。本研究では、発がんに至る腸内環境全体の変動がもともとの腸内環境に依存しているという仮説を立て、大腸がんに至る腸内環境の状態遷移モデルを腸内環境のタイプ別に構築することを目的とする。

Outline of Annual Research Achievements

近年、大腸がんへの腸内細菌の関与について多くの研究報告がなされている。一般に、腸内環境の群集構造は個人差があることが知られているが、これまでの報告では、大腸がんへの関与が、個人が持つ腸内環境の多様性にどのように依するのかという問題は未着手であった。本研究では、発がんに至る腸内環境全体の変 動がもともとの腸内環境に依存しているという仮説を立て、これまで蓄積してきた腸内環境の大規模コホートデータを入力情報として、大腸がんに至る腸内環境 の状態遷移モデルを腸内環境のタイプ別に構築することを目的とした。
本研究室ではこれまで、国立がんセンターと共同で大腸がん患者さんから収集した便検体や「生活習慣などに関するアンケート」を元にメタゲノム解析データやメタボローム解析データを大規模に蓄積してきた。本研究では、このデータを元に、異なる腸内細菌群集構造のタイプ間で、年齢、性別、BMI、および、喫煙習慣について比較をした。その結果、Prevotella属を主な構成種とするタイプには男性が多く、喫煙習慣を表すブリンクマン指数も高いことがわかった。また、30歳未満の被験者は全て Bacteroides属を主な構成種とするタイプの群集構造を持つことがわかった。これらの結果は、サンプルを年齢や性別ごとに層別化した上で大腸がんと腸内細菌の関連性を解析する必要を示唆している。そこで2022年度は、年齢を層別化し、特に若年性の大腸がん患者の腸内細菌叢に注目して解析を行なった。若年生の大腸がんは世界的にもこれまであまりサンプル収集が行われて来なかったため、知見が少ない。本研究では、健常者と大腸がん患者の腸内細菌叢を若年層とそれ以外で比較し、若年層特有の大腸がん関連腸内細菌を特定した。
