Outline of Research at the Start |
アスベストは、肺ガンや悪性中皮腫などを引き起こす。申請者はアスベストに結合するタンパク質を発見し、アスベストの検査方法に応用してきた。検査方法を世界標準化するためには、タンパク質の特異性をさらに向上させる必要があった。アンチフリーズタンパク質(AFP)は、氷に結合するユニットが繰り返した構造を持つ天然の無機結晶結合タンパク質である。本研究では、AFPの繰り返しユニットの変異ライブラリーを構築し、進化工学を利用することで、これまでにない高い特異性を持ったアスベスト結合タンパク質を作製する。本技術を開発することで、固体表面を高度に認識するタンパク質を創製する新たな方法論を確立する。

Outline of Final Research Achievements |
Asbestos causes lung cancer and other diseases. Technology is needed to easily detect asbestos at demolition sites. Antifreeze proteins specifically bind to the surface of ice. We succeeded in mutating this antifreeze protein and modifying it into a protein that binds to the surface of asbestos. The new asbestos-binding proteins not only binds strongly to asbestos, but also binds exclusively to asbestos, making it applicable to highly accurate asbestos detection technology.

Academic Significance and Societal Importance of the Research Achievements |
アンチフリーズタンパク質を利用して、高度にアスベストを認識するタンパク質を作製することに成功した。この新しいアスベスト結合タンパク質を蛍光で標識することで、アスベストを蛍光で光らせることで迅速にアスベストを検出することができる。この検出技術は、迅速にアスベストを検出する必要性がある解体現場や被災地などで活躍できると考えられる。また、アスベスト以外の無機物に応用できるため、有害な無機物質の検査に利用可能である。
