Outline of Research at the Start |
真核生物の複雑な細胞構造を形成するには、膜タンパク質を選別して適切な細胞膜ドメインへと送る極性輸送が必要である。申請者らは、膜タンパク質選別の中枢であるトランスゴルジ網(TGN)とリサイクリングエンドソーム(RE)が付着しており、接触と分離を繰り返していることを見出した。さらに、TGNとREの境界に局在するARFGEFの特異的阻害により、ゴルジ体とREの凝集体が形成されることを見出した。本申請では、このTGN局在ARFGEFを手がかりにして、TGNとREの接着、積荷タンパク質の選別・輸送、さらにポストゴルジ輸送小胞形成との関連を解析し、極性輸送の分子機構の解明を目指す。

Outline of Final Research Achievements |
Polarized transport of integral membrane proteins is an indispensable basis of highly developed structure of eukaryotic tissue. We have reported the two centers of protein sorting, trans-golgi-network (TGN) and recycling end-some (RE), are repetitively interacting. In this work, by the combination of genome-editing and pharmacology, we developed a system to regulate TGN-RE interaction, quickly and reversibly. In response to the removal of inhibitors, post-TGN transport to multiple directions was immediatly restarted.

Academic Significance and Societal Importance of the Research Achievements |
多種の疾患の原因として細胞内輸送の異常が同定されており、細胞内輸送のネットワークを分子レベルで明らかにすることは診断・予防・創薬の為に重要である。本研究では、トランスゴルジ網ーリサイクリングエンドソーム間の輸送についてこれまで知られていなかった新たな経路を明らかにするととともに、その薬理学的制御を可能にした。これにより細胞内輸送経路に関わる分子の探索が容易になり、新たな創薬ターゲットの発見につながると期待される。
