Outline of Research at the Start

本研究では、高い再現性・操作性と豊富な知見を兼ね備えた細胞性粘菌と好中球細胞を 対象に、微小流路を利用したハイスループットイメージング解析によってアクチンとその制御因子について包括的な観察を行う。機械学習や数理モデリング解析結果を生細胞観察実験と比較し、フィードバックさせることから、細胞遊走動態における多様な細胞行動の決定要因や支配法則の解明に挑む。

Outline of Annual Research Achievements

遊走性細胞の走化性の運動特性についてのルールの解明を目指し、本研究では、フォトリソグラフィを用いた微細加工技術と共焦点顕微鏡観察系の融合による、ハイスループットライブイメージング計測系を構築し、細胞遊走における細胞の個性とその特性を解析した。これまでの細胞遊走の先行研究の多くでは、せいぜい10から数十細胞分の遊走動態を解析していたのに対して、本計測系では、1000を超える細胞についての膜変形や細胞内分子動態解析を実現し、深層学習を利用した細胞輪郭の自動検出によって自動的な画像処理を行えるようにした。それらから、粘菌アメーバと好中球様培養細胞株HL６０の移動速度と細胞極性動態が細胞サイズに依存することを見出した。細胞サイズが中程度の場合、移動速度と細胞サイズは正の相関を示し、F-actinの細胞局在動態から評価した細胞極性は単一の方向、誘引物質濃度の高い方向に安定していた。ある一定サイズを超えた大きい細胞では、細胞サイズが大きくなるにつれてむしろ移動速度は減少し、細胞は誘引物質濃度の高い側のみならず低い側に先導端を形成し双極的な極性を示した。細胞サイズが小さくなると、先導端が一方向に安定せず誘引物質の高濃度側と低濃度側の間を振動し、その結果、方向的移動が見られなくなった。細胞の誘引物質刺激への応答性と極性動態の関係をさらに調べるため、マイクロ流路を用いて誘引物質方向の時間反転実験系による誘引物質方向を反転させた場合の方向転換動態解析から、細胞の発達段階に応じて濃度増加・減少に対する細胞反転動体が異なることを見出した。それら細胞では、アクチンバンドルの集積の仕方や推進力の起源が異なっていることがアクチン関連分子の集積パタンや時間空間動体から示唆された。
