Outline of Research at the Start

ヒト小脳では巨大な後葉半球部が、体性運動機能よりも小脳性高次機能障害や小脳性自閉症に関連する非運動機能に関与することが、近年、臨床症状や画像解析から示されている。しかし、その神経回路メカニズムの詳細は不明である。齧歯類の小脳を用いてヒトの小脳機能を解析するための小脳構造の相同性を明らかにしてきた我々のこれまでの研究成果に基づき、マウスを用いて、単一軸索投射解析、Aldoc-Venusマウスでの両方向性蛍光トレーサーによる高効率神経回路解析、神経回路破壊後の行動解析、形成発達機構の解析、スライスパッチ電気生理学的解析により系統的解析を行い、小脳の非運動機能の神経基盤を解明することを目指す。

Outline of Annual Research Achievements

ヒト小脳で巨大な体積を占める後葉半球部の小葉(第I脚、第II脚)は、臨床症状や画像解析から、体性運動機能よりも非運動機能に関与することが知られている。このことの神経回路基盤を理解することが重要であるが、そのためにげっ歯類モデルを用いて研究する場合、げっ歯類小脳では、半球部中央の第I脚と命名された1つの小葉のみがヒトの第I脚・第II脚に相当することをわれわれはこれまでに明らかにした。それに基づき、マウスを用いて、精緻な手法(単一軸索投射解析、Aldoc-Venusマウスでの両方向性蛍光トレーサーによる高効率神経回路解析、マウス行動解析、形成発達機構解析、電気生理学的解析)による系統的解析を行い、小脳の非運動機能の神経回路基盤を解明するのが本研究の目的である。
単一軸索投射解析、形成発達機構解析、電気生理学的解析、Aldoc-Venusマウスでの両方向性蛍光トレーサーによる高効率神経回路解析などを大学院生で分担し、2023年度までに、大脳から橋核を経由して小脳へ至る大脳小脳連関の部位対応的投射の構築の原則を解明した。大脳の連合野・辺縁系は橋核の吻側・内側を経由して、視覚野と聴覚野は橋核の外側を経由し、島皮質は橋核の外側を経由し、いずれも主として小脳の第I脚、第VIbc-VII小葉、および傍片葉に投射し、小脳の非運動機能の基盤となることが見いだされた。一方、大脳の運動野と体性感覚野は橋核の中央と尾側を経由して小脳のそれ以外の小葉に投射し、そこで、三叉神経核などを経由する末梢からの感覚入力と同じ小葉に収束することが判明した。小脳の第I脚は、小脳核の腹側に特異的に投射することも判明した。以上のように、小脳の非運動機能の神経回路基盤に関して新しい知見を得ることができた。

2: Research has progressed on the whole more than it was originally planned.

Reason

2022年度末に立てた繰越の2023年度の研究計画に含まれるほとんどの部分を予定通り進展させることができた。研究代表者を責任著者とする2編の論文を国際誌に発表することができ、英文書籍の1章の著作を発表することができた。これらの成果は、研究代表者と研究代表者の指導する大学院学生とで主要部分を担当した研究である。研究が進展した理由としては、留学生を含めた大学院生・研究員5名が本研究に参加してくれたこと、彼らの指導が順調にできたことが挙げられる。2021年度から設置されてマウスの一時的飼育ができる実験室(第二種実験室)を2023年度も引き続き利用できたことが本研究の進展に大きく寄与した。また、研究手法自体が、あまり挑戦的な新規なものではなく、これまでの研究代表者の開発してきた研究手法やマウス系統を発展的に引き継いだものであることも理由に挙げられる。さらに、所属大学と所属研究室の基本的研究環境が整っていたこと、昨年度までのうちに大学院生の力量で順調に研究成果を挙げられる態勢を整えてきたことなどが挙げられる。

Strategy for Future Research Activity

研究代表者は、東京医科歯科大学を2023年度末に定年退職するため、当初は、本研究も2023年度末で終了と計画していた。しかし、2024年度以降、3年または5年間、大東文化大学で常勤の教員職を勤めることとなり、本研究の延長が可能となった。大東文化大学への異動により、研究環境は大きく変わることとなる。実験室の整備には時間がかかることから、まずは、2023年度までに得られたデータの取りまとめと論文の執筆に注力することとし、次いで、2023年度までに得られた蛍光顕微鏡標本や明視野標本の観察、写真撮影および解析を行う。そして、組織学実験室が整備された段階で、2023年度までに得られた脳サンプルからの切片作成と免疫染色による標本作成等によって、研究を進展させていく。この脳サンプルには、脳・橋核・小脳投射パタンの大脳からの経シナプスアデノ随伴ウイルス(AAV)による標識の実験で得られたサンプルも含まれている。動物実験室については、将来的に使えるように準備を始めておく。
具体的には、次の解析項目のうちのいくつかを完成させる。未発表の脳幹のいくつかの神経核(特に、橋被蓋網様核)から小脳への苔状線維投射パタンについての単一軸索投射パタンの解析、小脳皮質の特定の小葉、特に傍片葉が小脳核、下オリーブ核、および下オリーブ核よりも上流の神経核(中脳間脳移行部など)との間に作る部位対応的投射の解析、小脳の小葉構造とゼブリン縦縞構造がどのように形成されるかという発達過程の解析、大脳・橋核・小脳投射パタンの大脳からの経シナプスアデノ随伴ウイルス(AAV)による標識での解析、などが候補である。それらの解析によって、小脳の非運動機能の神経基盤をより明確に示していく。
