Outline of Research at the Start

コレステロールのエステル化酵素であるACAT1選択的阻害剤K-604が、脳特異的オキシステロールにより誘導される神経細胞死を抑制することや、アルツハイマー病(AD)発症に関わるアミロイドβ産生を減少させることを明らかにしてきた。そこで本研究では、細胞死やアミロイドβ産生に対するK-604の詳細な阻害機構の解明を目指し、我々がこれまで樹立してきた方法を用いて解析する。またオキシステロールが引き起こす非典型的な細胞死誘導機構について小胞体ストレス応答という観点から明らかにすることを目的とする。本研究によりK-604のドラッグリポジショニングとして、前臨床試験に進むための基盤研究を行う。

Outline of Final Research Achievements

In this study, we found that the brain-specific oxysterol 24S-hydroxycholesterol (24S-OHC) activated the integrated stress response, resulting in induction of cell death in a 24S-OHC esterification-dependent manner. We also found that acyl-CoA: cholesterol acyltransferase 1 (ACAT1) selective inhibitor K-604 induced degradation of C99, which is a substrate for amyloid β (Aβ)-producing enzyme, thereby reducing Alzheimer's disease (AD)-related Aβ production.

Academic Significance and Societal Importance of the Research Achievements

アルツハイマー病の根本治療薬は現在でも開発は成功しておらず、世界的な問題となっている。ACAT1の選択的阻害剤であるK-604は、これまでのアプローチと全く異なる新たな視点から、アルツハイマー病態に関与するAβ産生の抑制、さらにオキシステロール誘導性細胞死の抑制という相乗的な効果をもつことが示され、発症を予防且つ改善する治療薬の開発を目指す点において有望な治療薬となる可能性をもつことが考えられた。また、学術的な知見としてオキシステロールが統合的ストレス応答を引き起こすことを明らかにした。
