Outline of Research at the Start

アセチルコリン受容体であるムスカリンM1受容体は、細胞表面に存在することで細胞外からアセチルコリンを受容し機能すると考えられてきた。しかし、我々は、M1受容体が細胞の内側にも存在し機能することを明らかにしてきている。本研究課題では、遺伝子編集による点変異導入により細胞内M1受容体を消失したマウスを作出し、その記憶学習への影響を解析することで、新しい認知症治療法開発に貢献することを目的とする。

Outline of Final Research Achievements

M1 musucarinic acetylcholine receptors localize not only cell surface but also intracellular compartments. In this research, we aimed to analyse functional roles of intracellular M1 receptors. As a result, we developed a new tool to visualize activation of intracellular localized M1 receptor. In addition, we found several candidate transporters which could transport acetylcholine into the cells. Co-expression of the candidate transporter led to activate intracellular M1 receptor by extracellular acetylcholine. Our results will contribute to demonstrate the intracellular cholinergic transmission in vivo.

Academic Significance and Societal Importance of the Research Achievements

アセチルコリンの受容体であるムスカリンM1受容体は、認知機能に深くかかわり、アルツハイマー病などの治療ターゲットの一つとしても知られる。しかし、このM1受容体が細胞の細部でも機能しうることはまだ一般的に知られておらず、その役割も明らかではない。本研究の成果として細胞内でムスカリンM1受容体が活性化されることを直接的に示す新しいツールを開発した。また、アセチルコリンを細胞内へ輸送するトランスポーター候補も同定した。トランスポーターは細胞内M1受容体の活性化を制御する新しい創薬ターゲットとなりうるものであり、認知症などの病態との関わりについても興味深い対象として今後更に研究の進展が期待される。
