Outline of Research at the Start

網羅的なヒト腫瘍の遺伝子解析から、既知のがん(抑制)遺伝子の変異に加えて、NRF2や関連因子遺伝子の変異が見出されている。興味深いことに、NRF2遺伝子変異は扁平上皮がんに集中しており、腺がんでは頻度が低い。これらの変異は転写因子であるNRF2を恒常的に活性化させて、腫瘍の悪性化に寄与する。本研究では、後天的にNRF2を欠失あるいは活性化させる新しい動物モデルを作製して、食道の発がん過程におけるNRF2活性化の果たす意義を解明する。新たに作出する動物モデルを活用して、食道がんの進展におけるNRF2の関与と機能貢献を調べ、今後の食道がん治療へ繋がるような分子メカニズムを提案する。

Outline of Final Research Achievements

We examined how Nrf2 plays roles in esophageal cancers developed by a chemical carcinogen, using two model mice that harbor acquired deletion of Nrf2 or Keap1 genes in esophageal epithelium. Partial appearance o Keap1-deleted cells in esophageal epithelium conferred DNA damage to Keap1-expressing cells, resulting in 4NQO-induced tumor promotion. Nrf2-addicted cancers are found approximately 30% of esophageal squamous cell carcinoma. This study is useful to understand development of Nrf2-addicted cancers.

Academic Significance and Societal Importance of the Research Achievements

Nrf2活性化がんは、食道がんのみならず様々な癌腫で見つかっており、がん形成におけるNrf2の貢献は興味深いテーマである。Nrf2活性化を賦与した細胞からはがんは形成されず、むしろ組織から排除される細胞競合が観察された。本研究から、Nrf2の活性化自体は細胞のがん化には寄与しないことが明らかになった。Nrf2の活性化はがん形成の過程で獲得されて、がん細胞の生存に働くことが考えられた。
