Outline of Research at the Start

人獣共通感染症であるレプトスピラ症の病原体レプトスピラの宿主持続感染機の解明を目的として,トランスポゾン挿入レプトスピラ変異体ライブラリーを作製してラット感染実験を行い,次世代シーケンサーを用いて腎臓に定着できないレプトスピラ変異体を網羅的に取得し,これら変異体を用いて各種のin vitro実験を行うことで,宿主持続感染に必須のレプトスピラ遺伝子およびその機能を明らかにする.

Outline of Final Research Achievements

Tn-seq using transposon mutant library of Leptospira interrogans serovar Manilae identified 129 mutants whose capacity to colonize kidney tissues of rats were reduced. When mutant strain RS00325 or RS01180 were inoculated with their parental strain to rats, the number of RS00325 and RS01180 mutants in blood at three days post infection was reduced to 1/1000 and 1/60 compared to the parental strain.

Academic Significance and Societal Importance of the Research Achievements

レプトスピラの宿主持続感染機構を解明することは,本菌が引き起こす疾患であるレプトスピラ症のコントロールのために重要である.本研究で得られた変異体の解析により持続感染に関わるレプトスピラ因子を明らかにすることで,それらをターゲットとしたワクチンや治療薬の開発も期待される.
