Outline of Research at the Start

本研究はCARに最適化されたがん細胞膜上特異的エピトープを認識するScFvの提示を行う事を研究目的にしている。この目標を達成するために申請者は以下の個別課題を設定し、これらを達成する事により目標に到達する。
1. 本研究の目的に合致する網羅的抗体セットの取得・選択と完備、2. 本目的を達成する評価系の完備と網羅的データセットの取得、3. アンメットメディカルニーズにかない、かつ有効な治療が望める対象がん種の決定、4. がん特異性が高く、対象疾患に対して広く一般性がある、もしくはがんの不均一性に対応出来るCARの提示。
これら個別詳細目標達成を通じて本研究の目的を満たす。

Outline of Annual Research Achievements

本研究はがん細胞膜上タンパクの“特異的エピトープ”に対する抗体を取得し、これによりえられる一本鎖抗体(single chain Fv:scFv)からキメラ抗原受容体(Chimeric antigen receptor:CAR)に最適化したScFv-CARの開発を行うものである。そして本研究を通じて有望なScFv-CAR分子提示のみならず、現在存在するがん創薬分野の必要要件である、広く一般性をもった薬効とがん全体にまたがる不均一性という問題のパラドックスの解決にもつながる知見を与えるとともに、細胞膜上ターゲットタンパクを利用した様々な新規がん分子標的化マーカー治療法やがん免疫治療法開発にブレイクスルーを与える事を目指した。
がん特異的抗原に対する抗体では無く、がん特異的エピトープに対する抗体を用いて“抗原単位ではなく、極所構造単位”に焦点をあてて今までに無いScFvを作製し、CAR-Tに最適な合目的高特異性ScFv開発を進めた。また抗体の選抜にあたっては、アンメットニーズに対応する新規CAR-T治療法の開発の為には安全性確保の為、正常組織や細胞での発現が無い事も視野に入れ抗体選抜を行った。抗体スクリーニングにおいてはヒト抗体遺伝子を出来るだけ網羅的に組み込んだファージ抗体ライブラリーを使用して、固形癌を含む広範ながん細胞に対する抗体取得を試み、がんの細胞膜上の特異的エピトープを通じたCAR開発を通じて新知見を明らかにするため、特異的エピトープを認識するScFvセットの完備を行い、そのScFv断片のCARへの適性を確認した。
最終的には、CAR-Tに使用する最重要抗体を胃がんにおけるEpCAM、大腸がん、胃がんにおけるEGFR、すい臓がんにおけるJAM1そして肝臓がん、ATLを対象としたIgSF4と決定し、その抗体断片の選抜を行った。
