Outline of Research at the Start

研究代表者らは、これまでの研究にて高血圧症による血管構造変化における鉄の関与、特に細胞内鉄取り込み受容体 トランスフェリン受容体1の関与を明らかにしてきた。本研究では、研究代表者らが発見した血管構造変化におけるトランスフェリン受容体1に着目し、胸部大動脈瘤の病態生理におけるトランスフェリン受容体1の関与について検討する。そして、胸部大動脈瘤に対する新規治療戦略の開発を目指す。

Outline of Final Research Achievements

Associations between a cellular iron transport protein, transferrin receptor 1 (TfR1) and cardiovascular diseases have been reported. We investigated the role of TfR1 in the e pathogenesis of thoracic aortic aneurysm using vascular smooth muscle cells (VSMC) specific TfR1 knockout mice. As results, we found that TfR1 in VSMC plays a role in the pathogenesis of thoracic aortic aneurysm.

Academic Significance and Societal Importance of the Research Achievements

高齢者人口の増加に伴い、胸部大動脈瘤患者数は増加している。これまでの胸部大動脈瘤の病態解明が行われているが、未だ不明な点が多く、その予防法は確立されていない。そのため胸部大動脈瘤患者の予後はいまだ不良であり、新たな治療介入法が期待されている。
本研究では、細胞内鉄取り込み受容体であるTfR1に着目し、胸部大動脈瘤の病態形成過程におけるTfR1の関与を見出し、TfR1が胸部大動脈瘤の治療標的になる可能性を基礎研究より明らかにした。本研究成果は胸部大動脈瘤の新規予防及び治療法開発に寄与するため、学術的意義、社会的意義は高いと考える。
