Outline of Research at the Start

アルツハイマー病やポリグルタミン病などの神経変性疾患では、異常構造変化および凝集した原因蛋白質の蓄積が神経変性を引き起こすという共通の発症分子メカニズムが考えられている。本研究ではオートファジー抑制分子Rubiconに着目し、そのオートファジー抑制作用を阻害する化合物をスクリーニングすることで、神経変性疾患に対する治療候補分子の同定を目指す。また、加齢性オートファジー不全の原因としてRubiconの関与が考えられたため、加齢組織におけるRubiconの発現制御と役割を検討することで、加齢性オートファジー不全のメカニズム解明に取り組む。

Outline of Final Research Achievements

This study focused on Rubicon, an autophagy suppressor, to elucidate the mechanism of reduction in autophagic activity with age. We analyzed post-translational modification changes of Rubicon during aging using Drosophila, which is suitable for analyzing aging in a short period. The results revealed that Rubicon undergoes different patterns of post-translational modifications depending on aging and sex.

Academic Significance and Societal Importance of the Research Achievements

オートファジーはアルツハイマー病などの神経変性疾患のみならず、生理的老化にも重要である。本研究で見出したRubiconの加齢および雌雄による翻訳後修飾変化は、Rubiconの機能変化にも関与することが予想され、さらなる検証を行うことで上記疾患や老化のメカニズム、および予防・治療法の開発に貢献する可能性がある。
