DICの本態である凝固活性化状態と微小循環障害、および炎症と血管作動性物質との関連について、TF&LPS誘発DICモデルの2種類のDICモデルを用いて究明する。 臓器障害や血管内皮障害の見られないTF誘発DICモデルでは、臓器障害が高度なLPS誘発DICモデルを凌駕する一酸化窒素(NO)の産生が見られるが、その産生機序を解明することで、DICにおけるNOの役割を明らかにする。また、炎症と凝固のクロストークを遮断する方法を検討する。

Outline of Final Research Achievements |
The significance of vasoactive substances was investigated using a rat model of disseminated intravascular coagulation (DIC). Even in the same DIC model, the LPS model showed severe organ damage, but the tissue factor (TF) model showed almost no organ damage. The reason for this is considered to be that the TF model has high fibrinolytic activation (thrombi is easily dissolved) and that the production of nitric oxide (NO), which is a vasodilator, is enhanced. Conventional DIC treatment has mainly been anticoagulant therapy, but there is a possibility that vasodilator regulators will be developed as new therapeutic agents.

Academic Significance and Societal Importance of the Research Achievements |
DICは、全身臓器に微小血栓が多発して、究極の血栓症とも言える病態である。DICにおける病態解析の手法や、治療法改善の研究は、そのまま一般的な血栓症(脳梗塞、心筋梗塞、いわゆるエコノミークラス症候群など)に対しても応用可能である。ラットなどの動物を用いてDICモデルを作成することは容易であり、今後もDICモデルを用いた研究は、広く血栓症全体に対しても診療レベルの向上につながる可能性が高い。
