Outline of Research at the Start

生活環境中の化学物質への曝露によって親の生殖細胞に生じたDNA損傷が子のゲノムに与える遺伝的影響を明らかにすることを目的とする。加熱食品に含まれる変異原物質であるアクリルアミドを雄マウスに投与し、雄性生殖細胞のDNA損傷(DNA付加体形成)および遺伝子突然変異を調べる。さらに、次世代個体のゲノム配列を解析し、生殖細胞系列突然変異頻度を測定して次世代影響の有無を調べる。

Outline of Annual Research Achievements

本研究は、遺伝毒性物質が生殖細胞にDNA損傷および突然変異を誘発し、次世代個体のゲノムに突然変異(de novo mutation)をもたらすリスクについて検討する。雄性生殖細胞のDNA損傷(DNA付加体形成)が次世代個体ゲノムの突然変異にどのように関与しているかを明らかにすることを目的とする。
加熱食品に含まれる変異原物質アクリルアミドを用いてトランスジェニックマウス遺伝子突然変異試験を実施した。雄gpt deltaマウス(8週齢)に300, 100, 30 ppmの用量で28日間飲水投与し、最終投与3日後(28+3d)および最終投与100日後(28+100d)に精巣および精巣上体を採取した。精巣からDNAを抽出し、DNA付加体(N7-dG-GA)量を測定した結果、28+3d群においてDNA付加体量は用量依存的に増加した。精巣および精子のgpt突然変異体頻度は28+3d群の300 ppm投与群で有意に増加した。
次世代個体ゲノムへの影響を調べるため、28日間飲水投与の3日後(28+3d)および49日後(28+49d)に無処理雌個体と交配し、次世代個体を得た。28+49d群の無処理群と300 ppm処理群、28+3d群の100 ppm処理群、陽性対象としてENU投与群について、各1家族(両親+子4匹)の全ゲノム解析を実施した。SNVの親子間比較を行い、次世代突然変異頻度を算出した結果、28+49d群の300 ppm処理群では無処理群と比較して次世代突然変異頻度が有意に2倍増加し、主にA:T塩基対の塩基置換変異の増加が見られた。また、28+3d群の100 ppm処理群では次世代突然変異頻度が有意に2倍増加し、G:C to A:T変異と数塩基の短い欠失変異が増加した。引き続き、生殖系列突然変異における変異原曝露時の精子形成ステージの影響を分析する。

Reason

雄gpt deltaマウスを用いたアクリルアミド28日間飲水投与実験を実施し、最終投与3日後(28+3d)および100日後(28+100d)の精巣および精巣上体を採取した。DNA付加体測定を行い、投与群の精巣においてDNA付加体(N7-dG-GA)量の用量依存的な増加を認めた。また、遺伝子突然変異頻度測定を実施し、28+3dの精巣と精子において、gpt遺伝子突然変異頻度の有意な増加がみられた。次世代個体ゲノムへの影響を調べるため、最終投与3日後(28+3d)および49日後(28+49d)に無処理雌個体と交配して次世代個体を得た。28+49d群の無処理群と300 ppm処理群、28+3d群の100 ppm処理群、陽性対象としてENU投与群について全ゲノム解析を実施し、次世代ゲノムのde novo変異を検出した。アクリルアミドを投与した雄から生まれた次世代個体のゲノムにおいてde novo変異の有意な増加を検出することができた。精子細胞時に曝露した精子(28+3d)由来の次世代個体および精原細胞時に曝露した精子(28+49d)由来の次世代個体の変異の特徴を比較し、変異原曝露時の精子形成ステージが生殖系列突然変異に与える影響について分析を行っている。研究はおおむね順調に進展している。

Strategy for Future Research Activity

引き続きアクリルアミド投与雄マウスの子のゲノムに生じたde novo変異の解析を行う。28日間飲水投与の最終投与3日後の交配群(28+3d)の100 ppm投与群、最終投与49日後の交配群(28+49d)の300 ppm投与群および対照群の家族サンプルを用いた全ゲノム解析によりde novo 変異を検出した。陽性対照としてENU投与群1家族について同様にde novo 変異を検出した。精子細胞時に曝露した精子(28+3d)由来の次世代個体および精原細胞時に曝露した精子(28+49d)由来の次世代個体におけるゲノム中のde novo変異の有意な増加を認めた。曝露時期の異なる投与群の子のde novo 変異スペクトルを比較し特徴を分析している。アクリルアミド投与によるDNA付加体形成、遺伝子突然変異、および次世代個体ゲノム変異を総合的に比較し、変異原物質の生殖細胞遺伝毒性と次世代突然変異の量的・質的関係について考察する。
