Outline of Research at the Start

腫瘍血管が不規則に開閉し、そこから薬剤が腫瘍組織へ勢いよく漏出するという現象(nano eruption)を発見した。現在この現象の機序解明に取り組んでいる。研究の過程でnano eruptionの直前に、血管壁外側に高輝度の顆粒が出現するという、全く新しい知見を得た。本研究では、顕微鏡の撮影条件を工夫してnano eruptionと高輝度顆粒の関係を詳細に観察する。

Outline of Annual Research Achievements

腫瘍血管が不規則に開閉し、そこから分子ナノDDSが腫瘍組織へ勢いよく漏出するという、極めて動的な現象(nano eruption)を発見した。現在この現象のメカニズム解明に取り組んでいる。研究の過程でnano eruptionの直前に、血管壁外側に高輝度のvesicleが出現するという、全く新しい知見を得た。 本研究では、この高輝度vesicleを詳細に検討するために1つのnano eruptionに着目し、空間的分解能(XYZ)と時間的分解能(T)を限界まで向上させる。高輝度vesicleの出現タイミングと血管壁との相対的な位置関係、血管が破綻する瞬間、破綻した血管壁の詳細な構造、薬剤が噴出する瞬間、噴出の初速、噴出の収束に伴う血管壁の状態など、従来の撮影条件では不可能であった微細な構造変化を観察し、nano eruptionのメカニズム解明に迫る。研究計画に基づいて、血管内皮細胞をはじめとするnano eruption周囲の細胞や構造を明確に描出する蛍光標識の条件、またnano eruptionの前後における高分子ナノDDSの挙動を正確に把握可能な撮影条件を検討した。
