Outline of Research at the Start

果物アレルギーは、子どもに多い卵や牛乳のアレルギーと異なり、成長してから発症し、さらには対象となる果物が増えていくという特徴を持っている。申請者は代表的な植物アレルゲンであるLipid Transfer Protein(LTP)の研究過程で、新規アレルゲンGibberellin Regulated Protein(GRP)を発見した。本研究では、両者を明確に分離し、アレルゲンタンパク質としての特徴を明らかにする。「GRPがなぜ重篤なアレルギーを引き起こすのか」、という謎を解くことが、難解な果物アレルギーの作用機序を正しく理解する近道であり、診断・治療・予防への道筋を開くと考えている。

Outline of Final Research Achievements

Gibberellin regulated protei (GRP) has attracted attention as an allergen that causes severe fruit allergy, but its details have not been elucidated. In this study, we clarified foods that GRP-allergic patients should be careful of by analyzing cross-reactivity using monoclonal antibody (mAb) techniques. Subsequently, we found a method for reducing allergenicity of GRP, which is known to cause symptoms even in heated fruit. Furthermore, by purifying various GRPs other than peach and apple, which have been successful so far, and preparing novel anti-GRP mAb, we have established a foundation for comprehensive immunological analysis of GRPs.

Academic Significance and Societal Importance of the Research Achievements

これまでの食物アレルギー治療は原因食品を食べない「徹底的な原因食物の除去」が主であったが、現在は食べられる範囲までは食べる「必要最小限の除去」が推奨されている。そのため、アレルゲンをタンパク質レベルで解析する「コンポーネント解析」は、効果的な治療や安全・安心な喫食のために欠かせない。本研究では、重篤な果物アレルギーの原因として注目されているGRPについて、GRPアレルギー患者が注意すべき食品を明らかにすることができた。また、今回見出したアレルゲン性を低下させる方法を応用できれば、GRPアレルギー患者が果物を安心して食べられる可能性を広げることができると考える。
