Outline of Research at the Start

我々は関節リウマチの新規治療標的を探索している。関節炎モデルマウスを用いた実験で滑膜線維芽細胞に発現するDNAメチル化促進因子Uhrf1(Ubiquitin-like, containing PHD and RING finger domain 1)を特異的に欠損させると関節炎病態が悪化することを見出した。本研究は関節リウマチにおけるUhrf1の分子機構をマウスとヒトサンプルを使用して明らかにし、新規治療標的を探索する。

Outline of Final Research Achievements

In previous study, we have tried to explore epigenetic regulator and found that Uhrf1 was significantly increased in arthritis tissue and expressed in synovial fibroblasts. Here, we revealed that UHRF1 expressed in synovial fibroblasts negatively orchestrate multiple pathogenesis such as Th17 recruitment and apoptosis resistance in rheumatoid arthritis and that Uhrf1 stabilization improve inflammatory arthritis pathogenesis using clinical specimens and model mice.

Academic Significance and Societal Importance of the Research Achievements

現在、関節リウマチ治療において生物学的製剤等が奏功しているが、未だ20-30%には十分な効果が認められない。本研究成果より、滑膜線維芽細胞に発現するUHRF1は様々な関節リウマチ増悪因子の発現を網羅的に抑制していること、UHRF1発現を維持すると関節炎病態が改善することを臨床サンプルやマウスモデルを用いて明らかにした。これらのことから、関節リウマチ患者におけるUHRF1発現維持は増悪因子を網羅的に抑制する新規治療戦略として期待される。
