Outline of Research at the Start |
① 培養ヒト細胞株に正常酸素または低酸素環境下でLET 20~120 keV/μmの炭素イオン線を照射し、DNA損傷マーカーを蛍光免疫染色する。 ② 超解像顕微鏡をもちいてマーカー分子の蛍光シグナルを3次元定量する。 ③ 定量データを機械学習をもちいて解析し、LETと酸素状況からクラスター損傷の誘導と修復動態を予測するアルゴリズムを開発する。

Outline of Final Research Achievements |
In the first fiscal year, we obtained high-precision 3D data of double-strand breaks induced by carbon-ions of various LETs under normoxic conditions. However, due to the subsequent COVID-19 pandemic, it became practically impossible to carry out further experiments (see the annual performance reports for details). Thus, we changed the research plan to focus on publishing all the data obtained up until that time, including secondary findings, in order to contribute to scientific community. As a result, we published three papers in English peer-reviewed journals and made nine presentations at scientific meetings.

Academic Significance and Societal Importance of the Research Achievements |
申請者の知る限り正常酸素環境下で多様なLETの炭素イオン線によって誘導される二本鎖切断の高精度3次元データの取得は世界初である。今後はこれを論文として発表する。発表されれば同データはLETの最適化による炭素イオン線がん治療のさらなる個別最適化に資する重要な生物学的基盤情報となると考えられる。
