Outline of Research at the Start |
本研究では、PA患者より外科的切除された副腎手術検体を用いて、Ca代謝を調節する因 子(PTH, ビタミンD, Ca等)の受容体の発現動態を病理組織学的に解析する。更に、ヒト 副腎皮質癌培養細胞を用いて、PTHやビタミンD、Ca濃度等のアルドステロン合成に与える 影響を機能的に解析する。Caの代謝機構がアルドステロン合成をどのように制御しているのかを解明する事を目的とする。

Outline of Final Research Achievements |
In this study, the effects of Ca ions and Ca channel blocker on steroidogenesis, especially for aldosterone biosynthesis in adrenocortical cells were histopathologically investigated and further verified by in vitro study. The present study has demonstrated a possibile new therapeutic target or a potential prognostic factor for secondary complications of osteoporosis in the patients with primary aldosteronism.

Academic Significance and Societal Importance of the Research Achievements |
本研究成果は、若年者に多く、治療抵抗性の高血圧症を来す原発性アルドステロン症患者における二次性のCa代謝障害や骨粗鬆症の二次予防を目的とする新規バイオマーカーの発見に貢献するのみならず、Caイオンのステロイド合成に与える直接的影響を明らかにすることでアルドステロン過剰産生に対するCa拮抗薬の治療薬としての可能性についても検討出来た。これらの知見は今後の原発性アルドステロン症の診療に影響を与える臨床的意義の大きいものである。
