Outline of Research at the Start |
脳に損傷が加わることで神経細胞死がおこり、それが様々な機序で成熟することでてんかん原性が獲得され、将来てんかんを発症することが知られている。しかし、てんかん原性獲得機序の解明を目指した様々な研究が行われてきたが、未だ解明には至っていない。そこで、痙攣発作により発現するマトリセルラー蛋白の1つであるテネイシンCに着目し、てんかん重積マウスモデルを用いて、テネイシンCの発現を抑制することで海馬の神経細胞アポトーシスが抑制され、さらにてんかん原性獲得を防ぐことができるか検討する。加えて、テネイシンCが神経細胞アポトーシス及びてんかん原性獲得を起こす情報伝達機構を解明する。

Outline of Final Research Achievements |
This study failed to demonstrate that a matricellular protein tenascin-C contributes to epileptogenesis via neuronal apoptosis. However, it is revealed that activation of platelet-derived growth factor receptor (PDGFR) caused neuronal apoptosis, possibly leading to the development of epileptogenesis, while a PDGFR antagonist inhibited neuronal apoptosis. As a PDGFR antagonist is clinically available, it may be easy to apply it to clinical practice. This study may enhance further studies, leading to the development of new anti-epileptic therapies.

Academic Significance and Societal Importance of the Research Achievements |
てんかん患者の約80%は現在の抗てんかん薬で良好にコントロールされるが、残りの約20%は治療困難であり、新たな抗てんかん薬の開発が望まれる。新たな抗てんかん薬を開発するためには、てんかんの発生およびその脳への障害機序を解明する必要があるが、難治性てんかんが神経細胞死を誘発する機序は未だ十分には解明されていない。本研究の主な成果は他疾患に対し臨床で既に用いられている薬剤が、てんかん原生獲得阻害に有効である可能性を示したことであり、まさしくドラッグリポジショニングによる創薬につながる研究として価値あるものと考えている。本研究がさらに発展し、新たな難治性てんかん治療につながる可能性を秘めている。
