Outline of Research at the Start

A:弾性線維欠損マウスを用いてポリープ状脈絡膜血管症(PCV)のモデル動物を作成し、発症病態を解明する。
B:PCVが観察できた臨床症例を集約し、患者背景及び蛍光眼底造影、光干渉断層計などの画像所見を詳細に解析することで臨床例におけるPCVの発症病態を解明する。
AとBで解明された発症病態に基づいてPCVに対する新規治療法を開発し、臨床例での治療効果の評価を行う。

Outline of Final Research Achievements

There were no significant difference about the formation, progression, and speed of choroidal neovascularization (CNV), progression with regards to wild and LTBP-4 gene knocked out (KO) mice. However, less power of retinal photocoagulation led to the formation of CNV in LTBP-4 KO mice compared to wild mice because LTBP-4 KO mice had the weakness of elastic fiber in Bruch's membrane.

Academic Significance and Societal Importance of the Research Achievements

弾性線維の脆弱性が脈絡膜新生血管をより生じやすいことがわかった。弾性線維形成にはさまざまなタンパク質が関与し、中でもLTBP-4ノックアウトマウスでより脈絡膜新生血管をより生じやすいことから、既存の抗VEGF薬に新たな成分、弾性線維脆弱化促進する蛋白をブロックすることで、脈絡膜新生血管の発生・活動性を減弱させる可能性がある。
