Outline of Research at the Start

シアル酸を含有する糖脂質(ガングリオシド)は細胞膜上でタンパク質やコレステロールとの集合体(脂質ラフト)を形成することによってシグナル伝達を担うと考えられている。その中でも、神経細胞に豊富なc系列と呼ばれる一群は、神経細胞の分化・増殖のみならず、アルツハイマー病への関与が示唆されるが、生物学的機能の詳細は未知と言ってよい。本研究では、c系列ガングリオシドの化学合成、さらには機能理解のための観察用・解析用プローブの開発を行う。これら合成分子を用いて脂質ラフトとの関係性を明らかにし、c系列ガングリオシドの機能の分子基盤を解明する。

Outline of Final Research Achievements

This study focuses on the membrane domains (lipid rafts) formed by glycolipids (gangliosides) in nervous tissue. Ganglioside analogs developed in this study revealed significant interactions of gangliosides to understand the functions of lipid rafts and their formation mechanisms.

Academic Significance and Societal Importance of the Research Achievements

神経疾患と深く関与する重要な生体分子でありながら入手が困難であった糖脂質を簡便に化学合成する方法を本研究で開発した。さらに、これを発展させることであらゆる生命現象を担う細胞膜ドメインと呼ばれる構造体の詳細な解析を可能にした。本研究の成果は、神経疾患の治療・創薬研究へ発展することが期待できる。
