Outline of Research at the Start

生体では少数細胞の分裂異常が引き金となりガンや発生障害などの疾病が惹起されるが、その原理は明らかでない。これは、生体の随意の細胞集団に分裂異常を起こし、その影響を細胞・個体レベルで追跡できる手法の欠如による。本研究では、光照射によって随意の細胞集団に局所的分裂障害を誘導できる新奇化合物と、ゼブラフィッシュ胚の個々の細胞挙動と形態形成を同時追跡できる顕微鏡システムを駆使し、分裂異常の規模に応じた子孫細胞の形質変化や組織形態・機能への影響を実験・理論の両面から解析する。この知見に基づき、分裂障害に起因する個体機能障害を回避、軽減し、病態制御に有効性をもつ介入法を予測・実証することを目指す。

Outline of Annual Research Achievements

細胞分裂の制御は、生命の継承維持に必須の現象であるが、生体で起こる少数の細胞の分裂不全が発生や個体機能に及ぼす影響についての定量的な知見は乏しい。本研究では、独自の細胞分裂の光制御技術を駆使し、ゼブラフィッシュ胚に様々なパターンの細胞分裂異常を誘導した際の個体機能への影響を解析し、少数細胞の分裂異常に対する生体の許容性や脆弱性を理解することを目指す。本年度は、前年度に引き続き局所光制御による細胞分裂異常の誘導法の最適化を進めるとともに、光制御系の高いスイッチ性と可逆性を活かし、個体発生の様々なタイミングで様々な時間間隔で分裂異常を誘導した際の胚への影響を詳細に観察した。この結果として、初期発生の各段階で、分裂異常への感受性が複雑に変化することが明らかになってきた。具体的には、受精後２時間のタイミングから１時間置きに分裂異常誘導のタイミングと期間を変更しながら胚観察を実施したところ、初期卵割期に数時間の規模で分裂異常を起こしたケースでは原腸形成から体節期にかけて胚が致死となるのに対し、同時期に数回分裂異常を誘導したケースでは、甚大な染色体分配異常や間期核形状異常が生じるにも関わらずほとんどの胚が咽頭胚期まで生存した。さらに初期卵割期に短期間分裂異常を誘導する場合にも、そのタイミングに依存して、咽頭胚期の形態に多様なタイプの異常が引き起こされることが分かった。また、受精後６時間を経過した後に全身的な分裂異常を数時間単位で導入した胚は、やはり甚大な染色体・核異常が見られるものの、ほとんどの個体で形態異常が視認されず、少なくとも数週間生存可能であることが分かった。これらの結果から、発生時期に応じて胚の分裂異常への許容性や、反応性が劇的に変化することが分かってきた。

Reason

前年度から引き続き実施している局所光制御による分裂導入は、条件最適化の途上にあり、計画よりも遅れがある一方、前項に詳述した時間的制御に注力した結果、胚の分裂異常感受性に、予期せぬ顕著な時期依存性があることを見出した。分裂異常が引き起こす個体レベルの表現型は、分裂異常のタイミングや時期が30分から1時間変化するだけで顕著に変化するため、本課題で使用している高いスイッチ性をもった光制御ツールを使うことで、はじめてこのような現象を捉えることができるようになったと考えられる。この、時期依存性に関する検証には想定以上の進展が見られたため、総合して上記の自己評価とした。

日印の連携に関しては、コロナパンデミック期に構築した盤石なオンライン連携体制により、日本側チームの学生も含め、実験技術面のサポートや指導をインド側研究者に直接受けて実験系の改良を進めるなどの有機的な連携でプロジェクトを進めることができている。2022年度には日印代表が共同責任著者の論文をプレプリント発表する段階にこぎつけており、現在は共同で論文改定にあたっている。

Strategy for Future Research Activity

分裂異常誘導の詳細な時間制御が功を奏し始めており、発生時期依存的な分裂異常への脆弱性変化について重要な知見が得られ始めているため、検証の最重点をこの問題に引き続き置いて、論文化を目指す。上記パンデミックへの対処から、インド側の研究解析技術の日本側チームへの効果的な導入に成功していることから、多くのタスクを滞りなく実施する連携基盤が構築できており、次年度もこれを継続する予定であるが、対面指導協力の必要な場面では随時研究者派遣による問題解決を図る。
