BLT2 is a G protein-coupled receptor (GPCR) which is activated by 12-hydroxyecotetraenoic acid (12-HHT). BLT2 is important for the maintenance of barrier function and wound healing in skin and intestinal epithelial cells. In this study, we aimed to analyze the structure of the BLT2 receptor and identify candidate compounds for BLT2 agonists through close collaboration with Heptares, a UK-based company known for drug discovery targeting GPCRs. Although we were unable to identify BLT2 agonist candidates, we succeeded in expressing large amounts of Gαi-fused BLT2 in Sf9 cells, and clarifying the structure of the apo form of the BLT2 receptor using cryo-electron microscopy. This structure will be useful to develop new BLT2 agonists for skin ulcer and inflammatory bowel diseases in the future.
