細胞毒素を利用した制がん研究への総合展開

Research Abstract

微生物由来の細胞毒素パラスポリン-2(PS2)が、特定の培養がん細胞やヒト摘出がんに対してがん細胞特異的に作用し細胞破壊を引き起こす。本年度、PS2の受容体に関する基礎研究と担がんマウスへの腫瘍効果や動態を評価した。昨年度、PS2に特異的に結合する細胞因子としてHep27タンパク質(Hep27p)を同定したが、Hep27p発現のノックダウンでのPS2感受性の低下は観察されなかった。またHep27pのPS2低感受性細胞への発現ではPS2の結合や細胞毒性は見られなかった。しかし、Hep27p発現低下がん細胞では細胞増殖が著しく低下しており、がん抑制遺伝子として機能している可能性を見出した。一方、PS2の立体構造情報を精査し、4箇所に各々蛍光標識反応を行った。このうち1種類の蛍光標識PS2分子が効率的に細胞に結合するものの毒性が低下することがわかった。そこで、がん可視化や標的運搬体としての生体分子としての作用を明らかにするため、蛍光標識PS2をマウス尾静脈より投与し、24時間後の蛍光シグナルを観察した。この結果、蛍光はがん部に優位なシグナルが観察されたが、他の主要器官にはほとんど観察されなかった。よって今回、がん識別能力のあるPS2プローブを得ることができた。一方、昨年度の研究結果の再現性を得るため、腫瘍モデルマウスに対するがん局部へのパラスポリン-2の注射投与を行った。KLN205、Colon-26ともに約24時間以内で劇的な腫瘍の縮小が観察された。対照投与群との比較では、パラスポリン投与群マウスで腫瘍抑制がみられた。
