Research Abstract

エームス試験などの微生物の試験では突然変異を誘発しないが,マウスでがんを誘発する化学物質(非変異発がん物質)が知られている。これらの物質のなかで,hydroquinone(HQ)の作用機構を出芽酵母を用いて調べた。HQはベンゼンの代謝物で,白血病などの血液系に作用する。その結果,Hog1タンパク質をリン酸化し活性化すること(MAPK経路の活性化),細胞周期をG2/M境界で停止すること,G2/M境界停止に特徴的なelongated budを形成し,その形成はSwe1(ヒトWee1のhomolog)タンパク質の安定化に依存することがわかった。Swe1の安定化はG2/M停止を導く。従って,HQはMAPKを活性化して,G2/M停止を導き,その結果異数性を誘発することがわかった。次に,酵母の結果がヒト細胞でも当てはまるかどうかを検証した。HCT116細胞に対して,HQは異数性を誘発した。HQはまたアポトーシスも誘発し,中心体の増幅も誘導した。一方,HQはG2/M期での細胞周期の進行を遅らせることも明らかとなった。HQはまたp53をリン酸化して安定化する。そこで,p53の役割について調べたところ,p53欠損株では,細胞周期の遅延が解消し,中心体増幅も観察されなくなり,異数性も観察されなくなった。P53正常株で,G2/M境界の移行を調節するWee1をノックダウンしたところ,HQによる中心体増幅も,異数性も観察されなかった。HQはp53を活性化するがODNA損傷に反応するATM/ATRの活性化は見られない。以上より,HQはヒト細胞ではDNA損傷を経ないで,p53を活性化し,その結果G2/M境界で細胞周期を停止(遅延)させることで,中心体の増幅を起こし,その結果異数性となる。HQのこのような作用は,OPPなどのエームス試験陰性化学物質の作用機構と同じであった。
