Research Abstract

DNAメチル化をはじめとするエピジェネティクな変化は、癌化において癌抑制遺伝子の不活性化などをとおし重要な働きをしている。申請者らはこれまでにマイクロアレイを用いたDNAメチル化の網羅的解析技術を用いて肺癌でメチル化の変化を示す遺伝子をみいだしてきている。これらにはmiR-29などをはじめmicroRNA(miRNA)がコードされる領域が多く含まれている。miRNAと癌との関連は近年、重要な話題のひとつであるとともに、それが小分子であることから治療への可能性を秘めた注目すべき研究対象である。しかしながら逆に不安定な小分子のRNAであるがゆえ臨床サンプルを用いた研究、診断の対象としてむつかしい。そこで本研究ではmiRNA自体ではなく、それをコードする安定なDNAのメチル化を対象として網羅的解析をおこなった。miRNAコード領域の上流25kbを含むアレイを用いて解析をおこなったところmiR-29をはじめ多くの領域で癌においてメチル化の変化がおこっていていることがわかり、癌におけるmiRNA解析の有用なツールとなることがわかった。
