Research Abstract

我々は、ストレス応答MAPキナーゼ経路(JNKならびにp38経路)の制御因子である新規MAP3K分子ASK2を見出し、ノックアウトマウスを用いた二段階皮膚腫瘍形成実験により、ASK2が腫瘍形成の抑制に働くことを明らかにしている。ヒトにおいては、多くの消化器系がんの細胞株、とくに食道がん細胞株においてASK2の発現が低下しており、食道がん原発巣における免疫組織化学染色によるASK2の発現検討では、約半数の組織において、周囲正常組織と比較してASK2の発現が低下していた。このような結果からASK2が食道発がんに対しても抑制的に働くことを予想し、マウスへの4-NQO投与による食道発がん実験を行った。その結果、野生型マウスにおいては多数の扁平上皮がんが形成されたのに対して、ASK2ノックアウトマウスにおいては形成された腫瘍数が少ないことが明らかとなり、予想に反して、少なくともマウス食道における化学発がん系ではASK2は発がんの促進に働くことが示唆された。さらに、マウスの食道におけるASK2の発現をin situ hybridizationにて検討したところ、ASK2はおもに粘膜上皮の基底細胞層に発現していた。ASK2発現細胞層は4-NQOの短期投与に応答して肥厚し、ASK2ノックアウトマウスにおいてはその肥厚が少ない傾向が認められたことから、ASK2は4-NQOに応答した細胞増殖に関わっている可能性が考えられる。以上の結果より、ASK2は、恐らくは組織や発がん刺激の違いに応じて、がんの抑制にも促進にも働きうることが示唆され、その機能を制御する機構を明らかにしていくことが今後の重要な課題である。
