Wnt亢進とBMP抑制の相互作用による胃がん発生機序の研究

Research Abstract

消化管上皮細胞の分化増殖はWntおよびBMPシグナルにより制御されている。Wntは未分化性維持に作用し、BMPは分化を促進する。また、BMPシグナルにはPI-3K/Akt経路の阻害によりWntシグナルを負に制御可能性も報告されている。遺伝子の変異やメチル化によるWntシグナル活性化は消化管腫瘍発生の原因となる。一方、BMP受容体I型遺伝子の変異などによるBMPシグナル遮断は遺伝性若年性ポリープ症の原因として知られている。しかし、胃腫瘍発生におけるBMPシグナル抑制の関与について、これまでにマウスモデルを用いた解析がなされていない。そこで、BMPシグナルの内在性阻害因子であるNoggin遺伝子を胃粘膜上皮で発現誘導させたK19-Nogトランスジェニックマウスを作製して解析した。K19-Nogマウスの胃上皮ではBMPシグナルが抑制されてSmad1/5/8のリン酸化が阻害された。しかし、K19-Nogマウスの胃粘膜には腫瘍病変は全く観察されなかった。これまでの研究により、胃粘膜でWntシグナルを亢進させても腫瘍は発生せず、WntとCOX-2/PGE_2経路を同時に活性化させると胃がんが発生することが明らかである。そこで、K19-NogマウスとK19-C2mEマウスを交配してK19-Nog/C2mEマウスを作製し、胃粘膜でBMP抑制と同時にCOX-2/PGE_2経路を誘導させて炎症反応を起こさせた。その結果、胃粘膜には大きな腫瘍が約20%の頻度で発生し、組織学的には若年性ポリープ症患者で認められる過誤腫に類似していた。したがって、BMP抑制は腫瘍発生の引き金となるが、それだけでは腫瘍発生に至らず、PGE_2経路の誘導に依存して炎症反応がともなう時に過誤腫が発生することがはじめて明らかになった。遺伝子発現解析の結果、WntとPGE_2で発生する胃がんとは異なる発現プロファイルが認められた。以上の結果は、今後の胃過誤腫の分子発生機序の研究に有用な情報となる。
