Research Abstract

DNAメチル化、ヒストン修飾が関与したエピジェネティック制御が、正常個体の発生・分化過程に重要な役割を果たし、その異常が様々な疾病の成因に関与していることが明らかとなりつつある。本研究ではHistone-GFPマウスを用いて、大腸非増殖分化細胞と増殖未分化細胞の分別に成功し、それぞれの遺伝子発現プロファイリングを行った。その結果、MycやCyclin Dなどのcanonical Wnt pathwayの標的遺伝子が増殖未分化細胞において高発現していることを確認した。機能的役割を明らかにする目的で、変異型β-cateninの発現誘導可能マウスを作製し、canonical Wnt pathway活性化による大腸腺管の分化増殖に対する影響を検討した。変異型β-cateninの強制発現により、大腸腺管のbudding、branchingが頻繁に観察され、新たにcryptが新生していることが示唆された。実際に、変異型β-cateninの強制発現により、大腸幹細胞、前駆細胞のマーカーであるLgr5, Msilの発現増加が確認され、canonical Wnt pathwayの活性化は幹細胞の増生を引き起こすことが示唆された。一方で、核にβ-cateninが蓄積した細胞では、細胞増殖のマーカーであるKi67の陽性率がむしろ少ないことが分かり,幹細胞の増生と細胞増殖能の亢進は必ずしも一致しないことが明らかとなった。今後、大腸の分化・増殖制御機構の破綻と、大腸がん発生の分子機構との接点を、分子病理学的なアプローチで解明していきたい。
