Research Abstract

RB1CC1はRBを介した細胞増殖抑制の系とTSC-mTOR-autophagyを介した蛋白合成・制御、細胞成長の系をクロストークさせながら、組織、生体の恒常的システム維持に貢献する。RB1CC1が細胞核内で、RB経路の転写へ貢献する機構について解析を行なった。
RB1CC1分子は細胞核内でhSNF5, p53両分子と転写因子複合体を形成する。本転写複合体はp16, p21,RB1すべての発現を亢進させ、RB経路を増強し、細胞増殖を抑制する。
RB1CC1の発現状態が乳癌症例の生存率に大きく関連していることを明らかにした。RB1CC1の細胞核内の局在を病理標本において簡便に判別することによって、臨床的予後が推定でき、治療選択の一指針にもなり得ることが明らかになった。RB1, p53の免疫組織化学的評価と組み合わせることによって、ER, PgR, Her2を用いたtriple-negative乳癌の鑑別に同等、もしくはそれ以上の精度で予後判定が可能となった。
一方で、精巣腫瘍特異的発現を示すDMNT3Lの生物学的機能、臨床学的意義を明らかにした。
