色素性乾皮症原因遺伝子産物によるゲノム損傷認識とその制御機構

Research Abstract

精製したヒストンアセチル化酵素(HAT) CBP/p300がUV-DDBと相互作用し、DDB2をアセチル化することを昨年度までに明らかにした。このDDB2の修飾部位を質量分析によって解析したところ、少なくともin vitroにおいてはN末端の40アミノ酸を含む領域に存在する7か所のリジン残基がアセチル化の標的となる可能性が示唆された。そこでヒト正常線維芽細胞WI38 VA13を親株として、このN末端領域を欠失した変異DDB2を安定に過剰発現する形質転換細胞株を樹立した。野生型DDB2の過剰発現により細胞の紫外線抵抗性が顕著に増強されたのに対して、N末端欠失DDB2を過剰発現する細胞株ではこの効果が明らかに減弱していた。一方、野生型DDB2は細胞の紫外線照射に伴ってCUL4リガーゼによるユビキチン化、およびプロテアソームによる分解を受けることが知られているが、N末端欠失DDB2は紫外線照射に応答した分解を受けにくいことが示された。以上の結果より、アセチル化部位として同定されたDDB2のN末端領域がユビキチン化の標的にもなっており、両者が共通のリジン残基を競合する可能性があること、またこの領域がDDB2タンパク質の安定性や細胞の紫外線感受性の制御において重要な役割を果たしている可能性が示唆された。現在、N末端欠失DDB2を含むUV-DDBを組換えタンパク質として精製し、in vitroでユビキチン化の基質となるかどうかを検討すると共に、7か所のリジン残基を非アセチル化体(アルギニン)、あるいはアセチル化模倣体(グルタミン)で置換した変異DDB2を作成し、その機能解析を進めている。
