Research Abstract

酸化と脱アミノ化は核酸,特にプリンヌクレオチドの生体内における化学修飾の代表的なものである。これらの修飾ヌクレオチドは元のヌクレオチドとは異なる塩基と対合する性質を有しており,突然変異ひいては発がんの原因となると考えられる。我々は,脱アミノ化プリンヌクレオチドが生体に及ぼす影響を解析するために脱アミノ化プリンヌクレオシド3リン酸を分解する酵素,ITPaseをコードする遺伝子のノックアウトマウス(Itpa遺伝子欠損マウス)を作製,解析を行った。Itpa遺伝子ホモ欠損マウスは成長遅延を呈し,生後14日前後で死亡した。その心臓は心筋症様の特徴を呈していた。生化学的解析より赤血球にITPの蓄積が,また複数臓器のRNAにイノシンヌクレオチドの蓄積が確認された。これらのことは,脱アミノ化プリンヌクレオチド(ITPなど)が細胞内で確かに産生していること,ITPaseはこれらの修飾ヌクレオチドを分解する役割を担っていることを示している。Itpa遺伝子ホモ欠損マウスはC57BL/6J genetic backgroundでは出生前に死亡する。このItpa遺伝子ホモ欠損胎仔の細胞のRNAには野生型胎仔の細胞のRNAに含まれる量に比べて50倍以上の量のイノシンが,またDNAには野生型胎仔の細胞のDNAと比べて約8倍の量のデオキシイノシンが蓄積していた。これらの胎仔から調整した初代Itpa遺伝子欠損MEFsは,野生型初代MEFsに比べて,倍加時間の遷延,染色体異常と一本鎖DNA切断の増加という表現型を示した。これらの結果を研究論文としてNucleic acids researchとCell death and differentiationに発表した。
