Research Abstract

私達は、白血病発症に関与する発がん遺伝子産物c-Mybに結合する因子を探索し、一群のFボックスを有する因子が、c-Mybに結合することを見出した。この内、Fbxw7はWntシグナル依存的なc-Myb分解に関与するが、Fbxw5は分解を阻害する事を見出した。Fbxw5の作用メカニズムを解析した結果、Fbxw5は核内でc-MybのSumo化を促進し、c-Mybによる転写活性化を阻害する事を見出した。
一方、私達は、c-Myb関連因子であるB-Mybが、クラスリン及びフィラミンと複合体を形成し、紡錘体形成に関与することを見出している。ショウジョウバエMyb(dMyb)がB-Mybと同様に、クラスリン及びフィラミンと複合体を形成し、紡錘体形成に関与するかどうかを解析した。そして、dMybの変異体を解析した結果、dMybは転写活性化能とは別に、紡錘体形成機能を有することが示された。さらに、dMybもB-Mybと同様に、クラスリンと複合体を結合して、紡錘体形成に関与することが明らかにされた。
また私達は、転写コリプレッサーSkiに関連する因子Snoの作用メカニズムを明らかにするため、Snoに結合する因子を解析した。SnoはSkiと同様に、Smad3/4と結合して複合体を形成しているが、Ski複合体には存在しない、新規の転写抑制因子を含むことが示された。
以上のように、がん抑制と発がんの両機能を有するMybファミリー、およびSkiファミリー転写因子群の制御メカニズムを理解するための知見が得られた。
