Research Abstract

酸化ストレスによるDNA損傷やシグナルは細胞の増殖・分化・細胞死の調節に重要な影響を及ぼし、一方、細胞側の抗酸化や酸化還元(レドックス)による調節は、活性酸素の消去による障害の防御のみならず、細胞増殖および細胞死の調節に関与している。細胞周期と細胞死調節に必須の役割を果たすレドックス調節因子thioredoxin (TRX)、ミトコンドリアに局在して細胞死調節に関わるthioredoxin 2 (TRX2)、および癌抑制因子的性格を持つthioredoxin binding protein-2 (TBP-2)/Txnipに注目して、癌における細胞システムの制御と破綻の機構をレドックスの観点から解析を行った。
今年度の成果は(1)thioredoxinは細胞周期制御蛋白cyclin D1の発現調節を行うことにより細胞生存調節を行っていることを明らかにした。さらに、thioredoxinはMAPキナーゼERKのリン酸化やその上流のキナーゼのリン酸化を制御し、MAPキナーゼシグナル伝達に重要な役割を果たすことを明らかにした。この結果は、thioredoxinシステムの制御により、癌細胞の増殖が抑制できる可能性を示す。(2)TBP-2の癌抑制作用の分子機構解析のため、TBP-2と相互作用する分子をプロテオミクス解析およびTBP-2の構造上の特徴から同定した。さらにTBP-2がインシュリンの分泌制御とインシュリンの感受性制御を行い、糖代謝制御に重要な分子であることを明らかにした。この結果は、TBP-2の分子機構を解析することにより、癌細胞がエネルギーを糖代謝に依存している機構を解明でき、癌増殖抑制への新たなアプローチにつながる可能性を示す。
