Research Abstract

オートファジーは、全真核生物が備える普遍的な細胞内大規模分解システムである。基本的には、細胞成分め代謝回転や飢餓などのストレス下での生存維持に働くが、近年の研究の急速な進展により病原細菌排除や神経変性抑制など予想外の多彩な役割を持つことが明らかになってきた。その中に発がん抑制がある。その根拠は、Beclin1というオートファジーに必須のタンパク質が、ある種のがんで発現していないことや、その遺伝子の片方を欠くBeclin1+/-マウスにがんが多発すること等である。
平成20年度の本研究において我々は、Beclin1、III型フォスファチジルイノシトール3キナーゼ、hVps15を共通成分とする複合体が少なくとも3種類存在し、共通成分に加えてAtg14Lを含む複合体がオートファゴソーム形成を促進し、UVRAGとRubiconを含む複合体がオートファゴソームやエンドソームがリソソームと融合する過程を抑制することを明らかにした。従ってBeclin1の発がん抑制能は、オートファジー促進の結果ではない可能性が出てきた。またオートファジーが逆にがん細胞のサバイバルに利用されている可能性について検討するため、オートファゴソーム形成を強力に阻害するAtg4B変異体を各種のがん細胞株に導入した。
