血中の微量なマーカー候補蛋白質・ペプチド探索と定量評価による新しい癌診断法の開発

Research Abstract

近年、プロテオーム解析を用いた癌の早期診断マーカーの開発の試みが数多くなされているがいまだに臨床応用に至った例はない。この主な原因として、血中のタンパク質の大部分を占めるメジャータンパク質(22種類で血清タンパク質の99%を占める)が邪魔になって微量なペプチドの検出が困難であることがあげられる。そこで本研究では、1.独自の手法を用いて血中の微量なタンパク質・ペプチドを抽出し、その中から癌の診断に有用な新しいマーカーを見つける。2.それらのマーカー候補タンパク質・ペプチドを定量的に測定する系を確立することで、癌の診断やスクリーニングに応用することを目的とした。
我々は昨年度、血中のabundantなタンパク質を除去することにより微量なタンパク質やペプチドを効率よく検出する方法を開発し、その手法を用いて、大腸癌患者血清中に特異的に発現する大腸癌組織由来の微量なペプチドzyxinの単離・精製・同定に成功した(J Proteome Res 2010 in press)。また、最新のプロテオーム解析技術を駆使して、種々の癌組織、特に大腸癌と乳癌組織中で癌特異的に発現増大するリン酸化タンパク質、膜タンパク質の探索を行い、癌の診断マーカーとなりうるいくつかのタンパク質・ペプチドを同定した。さらに、翻訳開始因子eIF4Hの2種類のアイソフォームのうち1種類だけがヒト大腸癌と食道癌の組織で発現が増大していることを見出し、その機能解析からそのアイソフォームが癌化に関わることを報告した(Int J Cancer 2010 in press)。一方、マーカー候補タンパク質・ペプチドを血液中で検出するために質量分析計を用いたペプチド定量法(MRM法)の開発も行った。以上の成果を用いて、これまで同定されたいくつかの腫瘍バイオマーカー候補をMRM法で血液中の検出・定量することによって種々の癌の早期発見が可能になると思われる。
