| Research Abstract |
低分子化合物ライブラリーをスクリーニングして同定した2種のSTAT阻害剤RJSI-1とRJSI-2について検討した。RJSI-1は活性化(リン酸化)したSTAT3およびSTAT5の核内移行を阻害することによってSTAT3/5の活性化を抑制する。一方、RJSI-2はSTAT3およびSTAT5のリン酸化、JAK2/1のリン酸化を抑制する。興味深いことにこれら2種の化合物はJAK2をはじめとする10種のキナーゼ(セリンスレオニンおよびチロシンキナーゼ)に対する直接の阻害効果がないことがキナーゼアッセイで確認された。また予備的的実験結果はこれらの化合物がSTAT3/5に対して直接結合しないことを示唆した。これらの結果はRJSI-1およびRJSI-2が新たなメカニズムでSTAT3/5を阻害することを示している。RSJI-1/2は担がんマウスモデルにおいてヒト骨髄腫細胞KMM-1の増殖を抑制することも確認された。現在、RJSI-1およびRJSI-2の改変を企業と共同で行っている。
