Research Abstract

本研究は、ウイルスゲノムを遺伝子工学的に改変して癌細胞で選択的に複製する単純ヘルペスウイルスI型(HSV-1)ベクターに直接免疫刺激遺伝子などの外来遺伝子を挿入し、ウイルス複製に伴う直接的な殺細胞効果に加え特別な機能を有する次世代の癌特異的増幅型ベクターの基礎開発を目的とした。治療域の広い増殖型第三世代HSV-1のG47Δを基本骨格として用い、BACプラズミドと2つの組換え酵素系を利用して、任意の外来遺伝子をHSV-1ゲノムに容易かつ的確に組み込むことができる独自の作製システムを活用した。腫瘍特異性の高い抗腫瘍免疫惹起型のHSV-1開発を目的に、可溶型マウスB7-1を発現するHSV-1(T-mB7.1-Ig)を作製した。HSV-1に感受性の高いA/Jマウスに、低免疫原性のNeuro2a神経芽腫細胞を植えた皮下腫瘍モデルで評価を行ったところ、T-mB7.1-Igは低用量の腫瘍内投与でも、対照ウイルスT-01に比べ高い抗腫瘍効果を示した。更に、基質依存的に発光するCBRルシフェラーゼを発現する遺伝子組換えHSV-1を作製した。CMVプロモータとHSV-1の後期発現型プロモータを用い、感染後すぐに発現が得られるタイプと、ウィルスの複製が開始してから発現が得られるタイプの2種類を作製したところ、前者のHSV-1(T-luc)の方が格段に高いルシフェラーゼ発現を示した。さまざまな経路でT-lucをマウスに投与した後にIVISで観察し、遺伝子組換えHSV-1の体内動態を追跡した。機能付加型HSV-1を活用し、さまざまながんに対するウイルス療法の治療効果向上と最適化を目指していく。
