Research Abstract

本研究は、がん組織の階層化に寄与すると考えられているがん幹細胞の維持・分化・増殖に関連したエピジェネティックな発現制御機構の解明と、その制御機構を標的とした新たながん治療薬の開発を目的として行った。
がん細胞には様々なエピジェネティクス異常が蓄積している。このうちDNAメチル化は安定した可逆性に乏しい修飾であるのに対して、ヒストン修飾による遺伝子発現制御には可逆性が保たれている。今回の研究では、固形腫瘍が腫瘍内不均一性を示す背景に、がん幹細胞の存在と、可塑性のあるエピジェネティックな制御機構、特にヒストンH3リシン27トリメチル化(H3K27me3)が関与していると考え、脳腫瘍幹細胞(Glioma initiating cell, GIC)を樹立して、その分化に関わるエピジェネティクス機構の解明を試みた。GICの分化誘導は培養条件により可逆的であり、分化誘導時にはポリコームタンパクのひとつであるEZH2(H3K27メチル化酵素)の細胞内局在の変化に伴う、ゲノムワイドなH3K27me3修飾変化が認められた。shRNAもしくは3-Deazaneplanocin処理によりEZH2の活性を低下させると、GICの分化誘導の阻害と、NOD-scidマウスでの腫瘍形成能が著しく低下した。一方DNAメチル化の変化はGICの分化誘導過程では観察されなかった。
可塑性を持ったがん細胞は、周囲環境に適応し増殖・浸潤すると考えられるが、その背景にはポリコームタンパク群によって誘導されるH3K27me3修飾による制御が重要な働きをしている可能性が示唆された。がん細胞の可塑性はがん組織の不均一性や多様性に関わり、腫瘍内に高い転移能・浸潤能を伴った細胞集団を形成することにつながる。がん細胞の可塑性を担うエピジェネティクス制御機構を標的とした治療薬の開発により、がん治療戦略の新しい展開が期待できると考える。
